From Mobile Genetic Elements to Cutting-Edge Genome Editors

Genome editing is revolutionizing biotechnology by providing high efficiency in genetic manipulation. Although basic platforms such as CRISPR-Cas system have laid the foundation for outstanding achievements, problems such as off-target effects have led to the search for enhanced approaches. In this chapter, novel technologies derived from MGEs are discussed: retron-based gene editing, especially Retron library recombineering (RLR), CRISPR-associated transposons (CAST), CRASPases, OMEGA systems, and Fanzor proteins. Because of their small size and high efficiency, they can be widely used in therapeutic purposes, demonstrating how improvements in the RNA-guided processes could contribute to the better understanding of the gene-editing technology. These pioneering developments collectively advance the function of genome editing and open new opportunities in the fields of therapeutics, agriculture, and food security, synthetic biology, and other areas of genetic engineering. This chapter offers extensive insights into the powerful next generation of genome-editing tools, focusing on their mechanisms as well as their viability and implications for the future.
