Application of PBPK Modeling in Formulation Development

The process of employing virtual drug development techniques, such as population pharmacokinetic (PopPK) and physiologically based pharmacokinetic (PBPK) modeling, in drug development is an approach that upon alignment of virtual and actual results to guide and accelerate actual drug development, thereby greatly improving the predictability and reliability of the outcomes in drug development. Software employed for modeling utilizes artificial intelligence (AI) to predict a drug's biological effects and pharmacokinetic properties, grounded in different principles. PBPK models, incorporating drug physicochemical properties, absorption, distribution, metabolism, and excretion (ADME) data, and dissolution data, can predict in vivo drug plasma concentrations and identify key parameters impacting pharmacokinetics. Drug formulations like oral medications, nasal sprays, topical treatments, and injections are tailored for specific administration routes to maximize effectiveness. Pharmacokinetics employs mathematical models to describe these drugs' kinetics in the body. Overall, PBPK modeling offers a comprehensive understanding of the interplay between various factors and formulations, facilitating formulation design, streamlining development processes, and enhancing the success rates of clinical trials.
