Characterization of the minor impurities during synthesis.of Alzheimer 8-protein

Synthetic Alzheimer 6-protein, JH O, was analyzed for minor impurities after assembly from activated esters of finoc-amino acids. Analytical RPHPLC show that 026-40 and 08-40 were the most common termination peptides. Co-ehrtion ofhctero-oli.gomeric protein species resuhed in band broadening on reversed-phase chromatography. Data on co-incubation of pure peptides indicate that a kinetic barrier had to be overcome for hetero-oligomeriz.ation. The presence of these peptides may affect the reproducibility of aggregation and toxicity assays.
