Bioassay

Emmanuel M. E. H. Lesaffre, Andrew B Lawson · 2012

Bioassay, short for ‘biological assay’, is a type of scientific experiment where a substance of interest is introduced to a living organism to assess the effects of the substance. This chapter focuses on the aspects of bioassay that arise mainly in drug-development environments. A preclinical in vitro mutagenicity experiment is carried out to assess the mutagenic potential of a new drug. The Ames/Salmonella microsome assay was developed to detect mutagenicity. A different assay arises when the focus is on changes in deoxyribonucleic acid (DNA) cell structure. The Mouse lymphoma assay (MLA) focuses on thymidine kinase locus mutation caused by base pair changes, frameshift and small deletions. All analyses were done with WinBUGS but could have been done also using SAS procedures. More specifically, the procedure GENMOD can be used to analyze the beetle data while the procedure Markov chain Monte Carlo (MCMC) is needed for the Ames assay. Controlled Vocabulary Terms Markov chain Monte Carlo estimation
