Review of drug repositioning of computational approach

Drug repositioning has been developing for decades, and it is becoming more and more popular in the world. Because it is less time-consuming, cheaper, and most importantly, the ability to identify new use of drugs to cure orphan diseases. There are many research papers on this concept, but there has not been very much of systematic review of the computational approach of drug repositioning, so this review is motivated. In this review, I analyze six different kinds of methods that affiliated to the computational approach. I identified that the drug-based approach is finding the quantitative relationship between chemical structures and biochemical activity. Nonetheless, it is restricted by the varied and complex relationships that associate a drug as an indication of a particular disease. The disease-based approaches, however, looking for similarities among diseases, are able to identify innovative drugs.
