Relativity between asymmetric nuclear division and strain degeneration in Volvariella volvacea

【Objective】The relativity between asymmetric nuclear division and strain degeneration was discussed to elucidate mechanism of strain degeneration in Volvariella volvacea. 【 Method 】 Three degenerated Volvariella volvacea strains were selected according to RAPD markers and colony morphology. The number and size of nucleus of a normal strain and three degenerated strains of Volvariella volvacea, PYb5, PYb6, PYb7, were observed and compared by nucleus staining. 【Result 】The results showed that many nucleuses exited in Volvariella volvacea hyphal cells. Bigger nucleus and smaller nucleus were produced by asymmetric division during mitosis in Volvariella volvacea hyphal cells.Compared with normal strain,the average number of nucleus per cell was less and the rate of asymmetric dividing nucleus in total nucleus is higher in degenerated strains. The average number of nucleus per cell of normal strain and three degenerated strains was 12.78, 10.06, 8.88 and 7.26. The rate of asymmetric dividing nucleus of normal strain and three degenerated strains was 2.5%, 10.5%, 15.3% and 23.7%. 【Conclusion 】Asymmetric nuclear division might be an important factor to cause strain degeneration in Volvariella volvacea.
