From Photons to Image: Data Models

Effective means of recording the images produced by a modern microscope are essential for recordkeeping and, even more importantly, for the analysis of experimental data. Technological advances over the past decades in detector development have played a crucial role in advancing the capabilities of modern microscopy. A wide variety of detectors with differing specifications are currently available. The specifics of the microscopy experiment dictate which detector should be used to allow for the best execution of the experiment. We introduce in this chapter some of the most important detector types and discuss their important properties. Particular focus is given to CCD, CMOS, and EMCCD detectors, the noise sources associated with these detector types, specifications such as the quantum efficiency and the readout speed for these detector types, and the measurement of some of the detector specifications.
