Drug discovery and development are among the most important translational science activities that contribute to human health and well-being. This chapter presents an overview of the drug discovery and development process from the laboratory bench through postmarketing approval. A brief history of drug discovery through the ages is included to provide perspective on the current situation and anticipated changes in the near future. Aspects of drug discovery include discovering drug targets, identifying and optimizing lead compounds, preclinical studies, and phase 1 through phase 4 clinical studies. New paradigms such as personalized medicine are also discussed. Numerous examples are included to illustrate various processes and principles.
