Effect of benzo(a)pyrene on Jun N-terminal kinase Activation of porcine aortic endothelial cells

Objective To study the effect of benzo(a)pyrene(BaP) on Jun N-terminal kinase(JNK) phosphorylation of porcine aortic endothelial cells. Method Porcine aortic endothelial cells were isolated and cultured in vitro. The cells were treated with BaP at 0, 0.1, 0.5, 1.0,5.0 and 10μmol/ L for 24 h,and the change of JNK phosphorylation was detected using Western-blot. Results The phosphorylations of JNK1/2 were both at low level without BaP stimulation, but the JNK1/2 was activated with the increasing BaP concentration, and the phosphorylation level was highest with BaP at 1μmol/ L, then decreased. However, the JNK2 was gradually activated with the increasing BaP concentration. Conclusion The JNK1/2 in endothelial cells was differently activated at different BaP concentrations, which may be one of the mechanisms of HSP 70 gene expression change and cell damage.
