Pharmacokinetics of metolazone tablet determined by LC-MS/MS in healthy volunteers

Objective: To study the pharmacokinetics of single and multiple oral doses of metolazone tablet 1 mg in Chinese healthy volunteers.Methods: Ten healthy volunteers were orally administrated with 1 mg metolazone tablet once a day for 6 days.The plasma concentrations were determined by LC-MS/MS method.The pharmacokinetic parameters were calculated by DAS 2.0 software.Results: The main pharmacokinetic parameters of metolazone tablet after single-dose(1 mg) and multiple-dose were as follows: Cmax were(20.6±4.8) and(22.4±5.0) ng·mL-1;Tmax were(1.60±0.6) and(2.4±1.4) h;AUC0~48 were(122.5±36.3) and(156.8±31.6) ng·h·mL-1;AUC0~∞ were(123.9±37.0) and(160.8±21.1) ng·h·mL-1;t1/2β were(7.9±1.2) and(8.9±1.4) h.Conclusion: No accumulation occurs after taking 1 mg of oral metolazone once a day for 6 days,and the dose regimen is well tolerated.
