Influence of VKORC1-1639G>A Genetic Polymorphism on Mean Daily Dose of Warfarin in Asian Populations:A Systematic Review and Meta-analysis

OBJECTIVE:To evaluate the influence of VKORC1-1639GA genetic polymorphism on mean daily dose of warfarin in Asian populations.METHODS:Related databases were searched by computer,and Meta-analysis was performed using Revman 5.0.23 software.RESULTS:16 articles including 10 in English and 6 in Chinese were included(n=3 016).There was difference in frequencies of VKORC1-1639GA among different population groups.Compared with VKORC1-1639AA group, mean daily dose of warfarin in-1639GG group,-1639GA group and-1639 GA+GG group increased about 113%[95% confidence interval(CI),(70%,156%)],52% [95% CI(30%,73%)] and 72%[95% CI(48%,96%)].Sensitivity analysis and subgroup analysis demonstrated that the results of Meta-analysis were stable and reliable.CONCLUSION:VKORC1-1639GA genetic polymorphism is significantly associated with the individual dosage of warfarin,and there is racial difference in the influence of it on clinical efficacy of warfarin.
