The inhibitory effect of euphorbia fischeriana stead on L 615 leukemia

Abstract Objective To study the potential anti leukemia effect of euphorbia fischeriana stead (EFS) on L 615 T lymphocytic leukemia. Methods The 80 purebred mice 615 were randomly divided into four groups: negative controls (Ⅰ), positive controls (Ⅱ), EFS 1 (Ⅲ) and EFS 2 (Ⅳ). Each mouse in negative controls received injection of physiological saline at left armpit, whereas those in other groups 10 4 RPMI 1640 culured L 615 cells respectively. 24 hours later the saline 0.3 ml (to Ⅰ), saline 0.3 ml (to Ⅱ), EFS 1 0.3 ml (to Ⅲ, equal to crude drug 0.036 g), EFS 0.3 ml (to Ⅳ, equal to crude drug 0.06 g) were given intragastrically daily for 7 days. On the 8th day, the 50% animals were sacrificed and the blood was taken, leaving the remainders for the calculation of ILS (increase in life span). Results The EFS 1 and EFS 2 had apparent inhibition on L 615 mice leukemia induced by attack of 10 4 cells, displaying the obvious prolongable mice ILS. The ILS of EFS 1 and EFS 2 group was 40% and 43% respectively, as conpared with controls. Due to the attack of L 615 cell the spleen of Department of Analytical Chemistry, College of Public Health, Shandong Medical University, Jinan 250012, China (Cui X and Liu P) Yan Tai Nursing School, Yan Tai 26400, China (Gong GM) Affiliated Hospital of Shandong Medical University, Jinan 250012, China (Du J, Wang BJ) mice in positive controls was significantly larger than those of negative controls, nearly increasing 3 times (P0.001). The spleen weights significantly decreased in ESF 1 and ESF 2 groups as compared to positive controls (P0.02), showing apparent relief. The significant difference of spleen weights between EFS 1 and EFS 2 was not observed. Compared with controls the body weights of mice groups in Ⅱ, Ⅲ, and Ⅳ decreased significantly, indicating little action of EFS on the recovery of mice body weights. The number of leucocytes in the blood of group Ⅱ was significantly larger than that in the blood of group Ⅰ, however, obviously decresed in groups Ⅲ and Ⅳ after administered EFS (P0.02), showing the effect of ESF on the inhibition of proliferation of leukemic cells and that enhanced with the increase of dosage. The results of the present research was coincident with the previous report that Chamejsme Sellera L., known as “Lang Du” as well in China, could inhibit the development of L 615 leukemia, indicating that the different genus of “Lang Du” could have the identical effects on L 615 leukemia. Conclusions The anti leukemia mechanism of EFS is not clear yet. After the administration of EFS the spleen swelling was relieved and the number of leukemic cells decreased, revealing that the anti leukemia effect might be carried out by enhancing the immune system and simultaniously inhibiting the proliferation of leukemic cells.
