CLONING OF ACC OXIDASE GENE OF CARNATION AND CONSTRUCTION OF ITS PLANT ANTISENSE EXPRESSION VECTORS

Chai De-yong · 2005

A pair of specific primers was designed according to the reported cDNA sequence of the DC-ACO(Dianthus caryophyllus L.ACC Oxidase),and a partial sequence of the gene was amplified from genomic DNA of a carnation variety 'Master' by PCR.Sequence analysis showed that the cloned and the reported sequences were basically coincident. An antisense expression vector, named pCAM-ACO_2,of the DC-ACO gene was constructed,in which the antisense sequence was controlled by the CaMV 35S promoter. The work laid the foundations of the future transgenic research for prolonging the flowering period of the transgenic carnation via antisense technology.
