Low-dose transdermal Lovastatin promote healing in the rat femur

Objective To explore the influence of low-dose transdermal Lovastatin on promoting healing in the rat femur. Methods 60 healthy,12-week-old female SD rats were chosen to establish the femur fracture internal fixation models. Lovastatin (5 mg. kg-1. d-1) mixed with promoting transdermal agent was applied to the drug-treated group,and the same dose of promoting transdermal agent was given to the vehicle group. External application of either drug or vehicle was used on the back of the rats after shaving for 5 days. Specimens were collected in 1,2 and 4 weeks after fracture,the fracture healing quality were evaluated by X-ray,bio-mechanics and histological analysis. Results Compared with the vehicle group,the fracture callus cross-sectional area in the drug-treated group increased 12.04% (P 0.01) and 14.55% (P 0.01) after 1 week and 2 weeks of fracture,and the fracture callus maximum load increased 39.37% (P 0.01) and 80.07% (P 0.01) after 2 and 4 weeks of fracture. In addition,the more neogenesis woven and compactly arranged bones were observed in the drug-treated group. Conclusions The data sug-gested that the low-dose transdermal Lovastatin could promote the fracture healing in rats.
