Antitumor Drug Sensitivity Test for K562 Cells by Resazurin Assay

To test the feasibility of resazurin assay in detecting the sensitivity of antitumor drugs,the sensitivity of three antitumor drugs( MMC,DDP,5- Fu) against human leukemia K562 cells was detected by resazurin assay and compared with conventional MTT assays. The results showed that the optimum concentrations against K562 cells measured by resazurin assay of MMC,DDP and 5- Fu were 100 μg /mL,30 μg /mL and100 μg /mL,and the optimum times were 6 ~ 16 h,10 ~ 24 h and 6 ~ 16 h,respectively. No significant differences between resazurin and MTT assays( P 0. 05) were observed. Therefore,resazurin method is consistent with MTT assays,and may be used for sensitive detection of antitumor drugs.
