Microplate-based screening method for quercetin antioxidant assay

Objective: To establish microplate assay model for screening antioxidant activity,using quercetin as research object,which had high antioxidant activity. Methods: The orthogonal experiment was designed according to L 9(3 4)orthogonal table after determining the single factor exploration. Results: The optimum experiment condition,which was A 3B 1D 2,that was,the concentration of DPPH was 200 μmol/L,the addition volume of DPPH was 175 μl and kept 20 min in room temperature,was determined by the direct-viewing analysis and variance analysis of the orthogonal experimental results.RSD of the evaluation test was 3.17%,which showed that the method was stable.And the standard of Trolox,PG,BHA and BHT were considered as control further certified the precision and feasibility of the model. Conclusion: The microplate assay is rapid,accurate,little sample,stable and reliable for the screening antioxidant activity of traditional Chinese medicine.
