Preparation and Identification of Inhibin α Subunite(1-26)-HRP Labeled Material

Inhibin α subunite(1-26) was coupled with HRP by EDC method,and then was isolated through sephadex G-50.The identification results showed that the D280 value obtained in the test increased with the increase of HRP concentration,suggesting that the enzyme had good activity.The activity identification curve of inhibin-HRP testing by ELISA indicated that the inhibin α subunite(1-26) purified material could be combined competitively with the antibody resisting inhibin.Among the tested 2 protein peaks,the inhibin α subunite(1-26)-HRP was in the washing liquid of one peak and that wasn't in the washing liquid of another peak.As the activity of enzyme and immunity of the linked complex was better in the test,it can be used for ELISA inhibin measurement strategy.
