Effect of Chinese Herbal Recipes for Supplementing Gan-Shen on Activating Mitochondrial ATP Synthase-6 Gene Expression in Mouse with Melanoma B-16

Objective To study the effect and mechanism of Chinese herbal recipess,single or composite,for invigorating Gan-Shen on the tyrosinase mRNA expression in murine melanoma cells.Methods Quantitative reverse transcriptional polymerase chain reaction(QRT-PCR)was used to analyze the effect of recipes on the tyrosinase mRNA expression in murine melanoma cells.The mRNA differential display reverse transcriptional pelymerase chain reaction(DDRT-PCR) was used to study the gene differential expression of B-16 murine melanoma before and after treatment with the recipes. Results Not all the recipes could increase the mRNA expression of tyrosinase,but the composite recipe and some single recipes could activate the special expression of mitochondrial ATP synthase-6 gene in mitochondria of mouse B-16 melanoma cells.Conclusion The therapeutic effect of Chinese herbal medicine could not be simply explained as the re- suit of increasing tyrosinase expression.There might exist other mechanisms.DDRT-PCR showed that the activated of ATP synthase gene expression is a possible key link.
