Oxidative stress and nitroglycerin tolerance: The effect of atorvastatin

Objetive To assess the effects of atorvastatin on the development of nitroglycerin tolerance during continuous transdermal therapy and to investigate the mechanism of its action. Methods 64 health male Sprague-Dawley rats were randomly divided into four groups, relaxation responses to nitroglycerin(9-10 mol/L或4-10 mol/L) were obtained, drawing the density-effect curve, testing the density value of 50% of its own maximal response. The activity of SOD and the contents of MDA were measured. Results The maximal relaxation responses in group atorvastatin+nitroglycerin (group Ⅳ) were significantly stronger than that in the control group (groupⅠ) (P0.01=. The density of nitroglycerin that lead to 50% maximal vascular relaxation in groupⅣ were significantly lower than that in the nitroglycerin group (groupⅡ) and groupⅠ(both P0.01=. The density-effect curve in group Ⅳ was in the left of groupⅡwhile in the right of groupⅠand groupⅢ(atorvastatin group). The activities of SOD in groupⅣ were significantly higher than that in groupⅡwhile the contents of MDA were significantly lower (P0.01=. Conclusion The result suggested that atorvastatin prevent nitroglycerin tolerance by depressing oxidative stress, although it could not be abolished completely.
