Identification and expression profiles of Cnaphalocrocis medinalis carboxylesterase genes

[Objectives] To identify carboxylesterase genes(Car Es) in Cnaphalocrocis medinalis and determine their expression profiles in various adult tissues. [Methods] C. medinalis transcriptome datasets were searched to identify Car Es. Bioinformatic software was used to analyze the identified Car E sequences, and a real-time quantitative PCR assay was performed to investigate their relative expression levels. [Results] A total of 15 Car Es were identified from the C. medinalis transcriptome datasets and named Cm Car E1-Cm Car E15. Of these, Cm Car E12 lacked the 3'-region whereas the other 14 sequences contained complete open reading frames(ORFs). With the exception of Cm Car E11, protein encoded by these Cm Car E genes showed features typical of Car Es, including a conserved pentapeptide, catalytic triad, and oxyanion hole. Phylogenetic analysis showed that the 15 Cm Car Es fell into different clades; Cm Car E13, Cm Car E14 and Cm Car E15 fell into the intracellular catalytic class, whereas the other 12 Cm Car Es fell into the secreted catalytic class. Cm Car E1, Cm Car E2, Cm Car E3, Cm Car E7, Cm Car E8, Cm Car E10 and Cm Car E13 were specifically expressed in the adult abdomen, whereas Cm Car E9 was specifically expressed in male and female antennae. Other genes were not tissue-specific. [Conclusion] Cm Car E1, Cm Car E2, Cm Car E3, Cm Car E7, Cm Car E8, Cm Car E10 and Cm Car E13 might possibly be involved in the metabolism of endobiotic and xenobiotic compounds, whereas Cm Car E9 might be involved in the degradation of odorant molecules.
