Study on Influencing Factors of Electrotransfection Efficiency

Objective: To explore the effect of influencing factors on electrotransfection efficiency. Methods: The mixture of CHO cells and DNA(plasmid carrying marker gene) were electrotransfected, then the effects of incubation temperature, osmotic pressure, electrotransfection parameter, cell cycle, etc. on cell survival and electrotransfection efficiency were investigated. Results: The results demonstrated that the incubation temperature didn't affect the cell survival, but could influence the electrotransfection efficiency. Hypotension condition, longer electrotransfection duration, pulse times could increase the electrotransfection efficiency. The cells at G2/M period were profitable to transfect. Conclusion: Electrotransfection efficiency and cell survival rate were influenced by temperature, osmotic pressure, electrotransfection parameter, cell cycle, etc. to some extent.
