Cytological ldentification and RAPD Analysis of Tritelytrigia Alien Disomic Substitution Line with Powdery Mildew Resistance

Dan Li · ACTA AGRONOMICA SINICA · 2003

Germplasm line Shannong 87074 551 with good resistance to powdery mildew, which derived from the progeny of Elytrigia elongatum and wheat, was identified by cytology and RAPD analysis. The results demonstrated that Shannong 87074 551 was an alien disomic substitution line between common wheat and E.elongatum, with chromosome number of 2 n =42, and its chromosome configurations was 21Ⅱ. Powdery mildew resistance of Shannong 87074 551 was dominant, and it was controlled by the chromosome of E.elongatum. Among 80 random primers used the specific DNA of E.elingatrm was only amplified by primer OPE13 from Shannong 87074 551, and also from the resistant individuals of F 2 segregation population between Shannong 87074 551 and Chinese Spring.
