Determination of captopril concentration in dog plasma by reversed-phase high-performance liquid chromatography

AIM The concentration of captopril in dog plasma were determined by high performance liquid chromatographic method. METHODS Captopril was stabilized by forming an adduct with p bromophenacyl bromide and the adduct formed was extracted with ethyl acetate benzene(1∶1),and then measured by RP HPLC using a ODS C 18 column as stationary phase and acetonitrile water glacial acetic acid(45∶55∶0 2)as mobile phase with ultraviolet absorption at 258 nm. RESULTS The method proved to be linear in the clinical range of 25~600 μg·L -1 and the lower limit of detection of captopril in plasma was 12 5 μg·L -1 . The RSD was under 4 95%( n =5)and 2 88%( n =5)for inter assay and intra assay respectively,the relative recovery rate was superior to 94 8%. CONCLUSION This method is fast,sensitive and validated for use in routine clinical analysis.
