BIOAVAILABILITY OF KETOPROFEN SUSTAINEDRELEASE CAPSULES

Using common ketoprofen(1) capsules and imported sustainedrelease 1 capsules as reference,the relative bioavailability of 1 sustainedrelease capsules made by our institute after single oral dose and its fluctuation in steady state after repeated oral doses in 8 healthy persons were studied by HPLC with naproxen as internal standard. The results showed that 1 sustainedrelease capsules made by us proved to have marked sustainedrelease property of pharmacokinetics and there was no significant difference in their absorption extent, the relative bioavailability was 85.4% and 101.9%, respectively. Given 100 mg×q12 h,the fluctuation of plasma concentration of 1 was much smaller than that of common capsules (50 mg×q6 h,50 mg×q8 h) in steady state,but its relative bioavailabilty was 102.3% and 84.5%, compared with the latter.
