Application of Hematoxylin stain method in determination of cell-mediated cytotoxicity

The aim is to establish Hematoxylin stain method for determination of cell-mediated cytotoxicity. Hematoxylin was employed to test proliferation of adherent Hep2 cells, cytotoxicity of human NK cell-lines against SGC7901 or Hep2 cells, and cytotoxicity of New Zealand white rabbit CTL against VX2 cells. We found that there was correlation between NK cell line cytotoxicity and effect-target cell ratio (E/T) within 10:1 to 1.25:1. Repeatability of Hematoxylin stain method exceeded that of MTT method. In rabbit CTL cytotoxicity assay, results from Hematoxylin stain method were consistent with that from MTT method. All theses results come to that Hematoxylin stain method is a specific, stable and cheap assay for determination of cell-mediated cytotoxicity of adherent cells.
