Effect of drug pair of Radix Rehmanniae and Cornus officinalis on the vascular of diabetic rats

Objective To detect diabetic rat serum nitric oxide(NO)/endothelin(ET),TXB2/6-keto-PGF1α,superoxide dismutase(SOD) and other factors the level,and investigate drug pair of Radix Rehmanniae and Cornus officinalis intervention in diabetic(DM) rats on vascular mechanisms.Methods High fat diet fed and STZ administered intraperitoneally once were used to made DM rats.Qualified DM rats were selected and treated with drugs intragastric administration for 12 weeks.At the end of 12 w,blood from rats were obstained after anesthetized with pentobarbital sodium and to make plasma and serum.Serum NO,T-NOS,and total SOD activity were detected and radioimmuno-assay was used to detect serum TXB2 and 6-keto-PGF1α and plasma ET levels.Results NO was decreased in DM rats and increased in drug pair group.There was opposite results in ET and SOD.Serum TXB2 was increased in DM rats and decreased after drug treatment,especially in drug pair group(P0.01),There was opposite results in 6-keto-PGF1α.Conclusions Drug pair of Radix Rehmanniae and Cornus officinalis can recovery NO/ET and TXA2/PGI2 balance,increase SOD activity,which indicates that drug pairs enhance the free radical scavenging capacity.Drug pairs can protect the vascular endothelium,inhibit a variety of vascular complications and development caused by diabetes including heart damage,aortic disease and nephropathy.
