Structural transform of dexamethasone and its inhibitory effect on proliferation of K562 cells

AIM: To transform dexamethasone's structure and to find the new bioactivity compound.METHODS: Structure modification of dexamethasone's side-chain was achieved by oxidization.The new compound was characterized by 1H NMR,13C NMR and ESI-MS spectra.Its inhibitory effect on proliferation of K562 cells was evaluated by MTT assay.RESULTS: Dexamethasone was reconstructed to a prospective new compound by oxidization.The new compound had a inhibitory effect on proliferation of K562 cells.At the concentration of 5 and 10 mg/L,the inhibitory rate was 9.64%±0.27% and 22.75%±0.35% respectively,it was higher than that of dexamethasone(P0.01).CONCLUSION: Dexamethasone is reconstructed to a new compound which has a more satisfactory inhibitory effect on K562 cells at low concentration.
