Effects of CV11974 on the myocyte viability

Objective To investigate the effects of a new non peptide angiotensin Ⅱ(AngⅡ) receptor (AT) antagonist CV11974, which is specific to the AT receptor subtype 1, on the myocyte viability and the cellular basis of myocardiocyte dysfunction, in order to provide the evidences for better management of heart failure and correct application of AT receptor antagonists. Methods Based on that the mitochondria of viable cells can act on methyl thiazolyl terrazolium (MTT) to produce formazan with light purple color, the effect of AngiotensionⅡ(AngⅡ) on the cultured myocyte viability and the intervention of CV11974 were investigated by means of colorimetric method. Results It was shown that AngⅡ caused a gradual increase in formazan optical density (OD) within 12 hours, which reached a peak at 12 hour, thereafter decreased, even became lower than the normal level at 24 hour. CV11974 successfully and significantly antagonized the effect of AngⅡ on the cardiac myocyte viability (CV11974 group vs ANGⅡ group, P0.05). Conclusion It is indicated that ANGⅡ can improve the myocyte viability at the early stage, CV11974 can effectively antagonize the action of ANGⅡ on the cardiac myocyte. The results have great significance for correct use of AngⅡ and efficient management of heart failure in the future.
