Cardiovascular diseases are the number one killer in the united state as well as developed western countries. According to estimates from the American Heart Association, about 80 millions of American people have one or more types of CVD. This is the condition despite of presence of many anti atherosclerotic drugs from long time. That’s why we have to move for other option that can cure the atherosclerosis. For that we opted for GSK-3 due to its role in inflammation apoptosis and lipid homeostasis. For this purpose model of advance atherosclerosis was developed and study was conducted on that rat model. Symptoms do not occur until a loss of at least 75% of the vessel diameter, most patients not presenting with less than 90% occlusions. We have used atorvastatin as standard along with ezetimibe for combination therapy while for test drug valproate was used due to its GSK-3 inhibiting activity. Valproate was not as effective as atorvastatin in controlling lipid profile. From biochemical indices analysis it is reasonably to conclude that we have to develop GSK-3 inhibitor which has higher lipid lowering capacity along with anti-inflammatory activity.
