Remarks on structural identification, modularity, and retroactivity

The “Modular Response Analysis” (MRA) reverse-engineering algorithm has been utilized in the systems biology literature in order to unravel the interactions between different “modules” in biochemical networks. We review the MRA technique, and discuss why its (mis-)application might lead to incorrect results due to impedance or load (“retroactivity”) effects, and present a computational criterion to discover stoichiometrically-dependent steady-state retroactivity.
