Activation of C-JUN N-terminal kinase (JNK) in mouse liver during hemorrhagic shock

Methods: Male mice were hemorrhaged to a mean arterial pressure of 25mmHg and sacrificed at 30 minute intervals from 0.5 to 2 hrs (3 animals per time point). Other animals were subjected to a mean arterial pressure of 25mmHg for 2.5 hrs., resuscitated with shed blood and crystalloid, and sacrificed at 30 minute intervals from 0.5 to 4 hrs following the initiation of resuscitation (3 animals per time point). Sham cannulated animals served as controls. Liver protein extracts were analyzed by Western blotting for JNK phosphorylation, as well as for the ability to phosphorylate its substrate, c-jun. Bands were quantified by densitometry, and statistically analyzed by ANOVA.
