Modified Osheroff-Ruffing: An Improved Trichrome Procedure for Glycol Methacrylate

Although the Osheroff-Ruffing trichrome procedure corrected many of the problems associated with the Masson trichrome stain in glycol methacrylate embedment, it produced its own problems and had a long turnaround time. We maintained the blue/red staining by modifying staining times and omitting the acid alcohol differentiation for Gill's hematoxylin. We also achieved a harder block and easier sectioning by increasing the embedding mixture ratio, decreasing the volume, and extending the polymerization process. Our modification gives consistently good results with a shorter turnaround time. (The JHktotechnol 10:249, 1987.)
