The survival kinetics of DBA/2 and CDF1 mice inoculated intraperitoneally with single L1210 leukemia cells were studied. This established a baseline of maximum expected survival for leukemic mice when no treatment was given. Leukemic mice were then treated with cyclophosphamide and amethopterin under conditions designed to reduce the number of cells surviving chemotherapy to a very low number (approximately one). The time-death distributions of mice dying with leukemia following treatment were compared to the distribution for the untreated single-cell controls. A considerable displacement (prolongation) of the survival curves for the treated groups was observed. This indicates the kinetics of this murine leukemia system are modified with some regularity by chemotherapy with the agents employed when treatment results in few surviving cells.
